Background
ThereactivityoftheantiserumisdirectedtotheFcsubunitsofthemajorisotypesofthehumanimmunoglobulinsystem.NoreactionisobtainedwithpurifiedIgG/Faboranynon-Igproteinofhumanserum,astestedbyimmunoelectrophoresisanddoublerADIalimmunodiffusion.Inimmunoelectrophoresisanddoubleradialimmunodiffusionusingvariousantiserumconcentrationsagainstnormalhumanplasmaandserum,thecharacteristicIgG,IgAandIgMprecipitinlinesareobtained.DirectimmunofluorescencestainingofcytoplasmicIgofappropriatelytreatedcellandtissuesubstrates;todemonstrateimmunoglobulinsorspecificantibodiesincellsandtissues;toidentifycirculatingantibodiesinSEROdiagnosticmicroBIOLOGyandautoimmunediseases;toidentifyaspecificantigenorimmunecomplexusingareferenceantibodyofhumanorigininthemiddlelayeroftheindirecttestprocedure.TheabsenceofactivitytothecommonIg/FabsubunitpreventsthereactionofthisconjugatewithimmunoglobulinsboundstoFcreceptorsonnon-lymphoidcells.Thisimmunoconjugateisnotpre-diluted.Theoptimumworkingdilutionofeachconjugateshouldbeestablishedbytitrationbeforebeingused.Excesslabelledantibodymustbeavoidedbecauseitmaycausehighunspecificbackgroundstainingandinterferewiththespecificsignal.Workingdilutionsareusuallybetween1:20and1:120.
